You can board the bus at any of the route's stops. The favored stops are Museo del Prado (Stop 1), Puerta de San Vicente, Plaza de Espana and Colegiata - Mayor
Walking tour: 3 Princess Street (Calle Princesa, no 3), past the hotel RIU Plaza de Espana, at the bus stop closest to Plaza de Espana

We bought the 48 hour passes for four people, and found this an excellent way yo move between the main tourist sites in the city. The staff were young multi-lingual and helpful. Well worth the money.
